Open Raman microscope
he Open Raman Microscope is based on Thorlabs Cerna range allowing customized construction of microscopes. Here we provide a base implementation of a non-confocal + confocal Raman microscope with 532 nm and 785 nm laser excitation. The system is modular with filter cubes and can be operated in a highly confocal setting for e.g., 3D cellular imaging or in a non-confocal (higher throughput) setting for rapid tissue imaging. Individual components can be replaced with alternatives as desired/required for the particular needs (assuming they maintain compatibility with all other components/software).
The basis implementation in the open sourced control software (ORM Controller) supports the following hardware:
Spectrometers/CCD:
Stages/movement control:
Widefield cameras:
For control of the microscope the following PC configuration is recommended:
Windows 10
Intel i9 Processor
64 GB Ram
Notes on modularity and customization:

Notes on modularity and customization:
- Any laser from the UV to NIR can be implemented given filters/lenses are compatible with the wavelength range.
- For any laser ensure that the laser is beam expanded to fill the back aperture of the microscope objective and this should be matched to the collection fiber coupling.
